The paper has far too much literature review and too little proposal.
The sections on in vitro model of oogenesis and photoactivation could likely be moved to the proposal section.
Looking into chromosome capture experiments, SNP-FISH,
Overall I think the topic would be good since its something significant and not understood. However I’ve found trouble getting experiments that would provide useful information. I had assumed that live cell or dynamic methods would be available for use in a cell culture system that would find useful and unique information regarding mechanism of genomic imprinting. The current experiments I had written up were not particularly well thought out since I wasn’t sure what useful or novel information they would provide. I’m trying to look at more assays that might be useful (TIVA, SNP-Fish, Chromatin Conformation Capture, ChIP-loop, forced looping, transcriptome analysis to look at the role of TADs, RNA fluorescence). I don’t know if you have thoughts on any of these. I have a final tomorrow but afterwards will start taking notes on methods and experiment ideas. Overall though the paper requires a sound experiment which would yield new and interpretable data so if you feel it is best to work with a different topic at any level (use different inspiration from Nature/Science, use the oogenesis system for different purpose, look at different aspect of genomic imprinting) I’m alright with going a different direction. But since the essay was my work then feel free to directly use any parts that might be helpful.

Please let me know what you think so far. Do you still want to use the in vitro oogenesis system? Do you still want to look at genomic imprinting? What ideas do you have currently?
I uploaded some miscellaneous notes I’ve taken today that I thought might be useful. If we are still looking at de novo genomic imprinting I feel a key part of this will involve using live-cell/dynamic methods that weren’t possible before and hopefully will yield useful info. Please let me know your thoughts so far so I know which direction to do research, ask my professor/friends etc.
