ANALYSIS OF THE INHERITANCE OF EYE COLOR IN DROSOPHILA In this multi-week experiment you will determine the inheritance pattern of an eye color mutant by performing and analyzing a series of crosses for eye color phenotype and presence of different pigments. Think about how this type of analysis compares with pedigree analysis for determining inheritance patterns. Although you will be working with a group to generate data, you will analyze your results and write up the first of two individual formal lab reports according to the guidelines at the beginning of this manual. The second formal lab report will be required for Ex. 8, UV Mutagenesis. Caveats: • The number of eye color phenotypes may be larger than the number you expect. Be observant! Failure to record the phenotypes correctly may make your data uninterpretable. • There is more than one way to set up this experiment. Work toward a conclusion that is consistent with your results. Do not be surprised if someone else’s (or an old lab report’s) results are vastly different from yours. Keep a careful record of your stocks and of your results. • Detailed record keeping is critical throughout this experiment; be sure to write everything down in your lab notebook. Week 1: Parental (P1) Cross Your lab group will have two initial stocks: one for females and one for males. To start this experiment, you will collect virgin females from the “for female” stock culture, and males from the “for male” stock. You will set up at least three vials, each with a male and a female, as indicated by your instructor. These are your parental (P1) crosses. Carefully record the phenotype and gender of the individuals that make up your parental cross and also be sure to note the mutant designation for each one, as indicated by your instructor. Week 2: Culture Maintenance Check cultures: transfer or rehydrate if necessary, remove parents, record observations and procedures. Week 3: F1 Analysis and Cross Record the results of the P1 cross: numbers of F1 flies of each eye color phenotype and gender combination. After consulting with your instructor, set up at least 3 vials of duplicate F1 crosses. Again, record the phenotype of each parent in the F1 cross. Week 4: Culture Maintenance Check cultures: transfer or rehydrate if necessary, remove parents, record observations and procedures. Week 5: F2 Analysis A. Analyze the F2 offspring for eye color and sex. Record all observations.
